For some reason smokers cannot seem to realize how obnoxious the stale smell of cigarette smoke really is. I smoked for 30+ years and have been quit for 7 years. I have been on both sides of that situation. I had no idea that I smelled so bad. Now I can get within 5' of someone and know if they smoke or not, simply by the smell. I have been car shopping and know if a smoker has owned the one I try out. Even walking in a cafe that allows smoking is a turnoff unless I am mighty hungry.
The place where I worked for 20 years was a very large factory. Had a cafeteria and two dining rooms, one for smokers on the west side of the cafeteria and one on the east side for non-smokers. In 2004 the plant was sold and the new company had a no smoking policy anywhere on the premises. The old smoking dining area was cleaned and the walls washed several times, and once even painted over the walls. The smell could not be eradicated. They finally locked it totally.
If your boss thinks a non-smoker will not know that the room was a smoker, then she is burrowing her head in the sand.
I am a peaceful sort but if that happened to me I would definitely raise some ruckus and contact the corporate headquarters.
