One drop of Lugol's solution contains about 6.25 milligrams of total Iodine content.

Adding one drop of Lugol's solution to a liter of water yields a solution that is about six parts per million Iodine by weight.

Adding one drop of Lugol's solution to six liters of water (about a gallon and a half), yields six liters of water, each liter containing one part per million of Iodine by weight.

For reference, city water that is fluoridated is often targeted to contain one part per million of fluoride, by weight.

Edit + HOWEVER, becuase the atomic mass of fluoride is about 19 and the atomic mass of iodine is about 127, one needs to have about 6.7 times as much weight of iodine than fluoride, to achieve the same number of atoms in such a solution. Stated another way, 19 grams of fluroide contains the same number of fluoride ions, as would 127 grams of iodine contain Iodide ions.

So, to produce a solution that has the same number of iodine ions that a 1 ppm (by weight) solution of fluoride has fluoride ions, one would need about 6.7 milligrams of iodine per liter. One drop of Lugol's solution in a liter of water therefore provides a solution that has almost as many iodine atoms, as a 1ppm (by weight) solution of flouride has fluoride atoms.
