A few people have talked about this in the Urine Therapy forum.
It seems like a common thing to do is poke the mole with a needle (make sure it's sterile!) or in some other way break or puncture the skin of the mole, and then apply the urine. This allows it to get into the blood throughout the mole much quicker. If you choose to experiment with this, I personally would suggest only going at it a little at a time, instead of gouging some big hole in the thing. :) However, doing it this way may possibly leave some scarring, depending on how much you poke... (although urine is reportedly very good for healing and preventing scars!)
Simply soaking without puncturing the skin should work too, but it may take a bit longer.

It should be permanent as far as that individual mole is concerned. If it actually is a mole and not a wart, anyway. Warts I think can come back unless you eliminate the "root" or whatever.
