Biochanin A Fomononetin Genistein Daidzein
Isoflavones are one of most characteristic classes of chemical compounds in higher plants. From a chemical points of view, they comprise a wide group of structurally related compounds with a chromane skeleton provided a phenyl substitute at C3 position. Natural isoflavones widely distribute in Legume family. However, the number and quantity of isoflavones are greatly different in different herbs. Red clover currently is considered as the herb which contains the most isoflavones. Biochanin A, Fomononetin, Genistein and Daidzein are mark active isoflavones. Specially, biochanin A is a major isoflavone extracted from red clover while it hardly exists in soy-derived extracts.
History
Red clover was originally used as a medicinal herb by native indigenous people of North America for whooping cough, gout and cancer. And traditionally, some people also used it in the treatment of asthma, bronchitis, coughs, and dermatophytosis , etc.
Natural Phytoestrogen
The isoflavones structures of red clover extract are similar with estrogen. Therefore, they can activate estrogen receptors(ER). Jonathan T.C. reported that product extracted from the leaves of red clover bind the ER effectively. The ER activity is contacted intimately with women estrogen level in body. Women will decrease estrogen level with age and cause related bone density problem. Charlotte Atkinson, enrolled women aged 49 to 65 years (N = 205) in a double-blind, randomized, placebo-controlled trial to test red clover extract for loss of Lumbar Spine Bone Density. The result is positive after 1 year administration of red clover isoflavones.
Activity on Menopausal
Peter et al reported that menopausal women treated with 80 mg isoflavones per day resulted in a significant reduction in hot flushes from baseline. At the end of the 12 weeks, there was a significant decrease in hot flushes of 44% between the active and placebo group, demonstrating the effectiveness of red clover extract in the management of hot flushes.
Activity on Cancer
In vitro studies showed biochanin A and genistein affect cellular expression of NADPH:quinone reductase in
colonic cells. The result suggests biochanin A and genistein may be implicated in colorectal cancer chemoprevention.
The study of prostate cancer cell LNCaP was cultured with Biochanin A indicated that biochanin A inhibits prostate cancer cell growth through induction of cell cycle arrest and apoptosis. Biochanin A-regulated genes suggest multiple pathways of action. Biochanin A inhibits the incidence and growth of LNCaP xenograft tumors in athymic mice.
Clinical experiment studied by Jarred
RA et al estimated intake of dietary isoflavones may halt the progression of prostate cancer by inducing apoptosis in low to moderate grade tumors.
Takahiko Gotoh et al reported that biochanina A is useful for rat mammaery carcinogenesis. Comparing the biochanin A-supplemented diet groups (50mg/kg) and control group, the incidence of rats with tumors was significantly decreased from32% to 80%.
Jih-Tay Hsu cultured the MCF-7 human
Breast Cancer cell line treatment with Biochanin A. Research data showed that biochanin A effectively inhibits human
Breast Cancer cell growth in vitro.
Kazuyoshi Yanagihara et al reported that biochanin A and genistein, inhibit the cell growth of stomach cancer cell lines in vitro. Biochanin A and genistein activate a signal transduction pathway for apoptosis. Moreover, in vivo experiments demonstrate that biochanin A can be used as an anticancer agent.
Antioxidant acitivy and improving immunity
Free radicals are very unstable molecules, which can react quickly with other compounds. Some free radicals arise during metabolism, and some are caused by environment pollution, UV-ray and cigarette smoking, etc. Free radicals can snatch electrons of normal cells continually, thereby damage living cells and accelerate cell decrepitude.
Experiment conducted by Wei et al showed genistein and daidzein significantly inhibit TPA-induced free radical formation. Furthermore, genistein is potent in inhibiting free radical generation. William banz reported that daidzein shows higher antioxidant level than SOD in liver cell of rat. Animal model experiments showed dietary administration of genistein for 30 days significantly enhances the activities of antioxidant enzymes in the skin and small intestine. Red clover extract is also potent in eraseing wrinkle, keep skin cell younger.
Zhang RQ reported that rat administrated formononetin or daidzein for 7 days significantly improves rat thymus gland weight and macrophage function. And studies on pigs showed fomononetin can also improve immunity. Schrepfer S reported that Biochanin A inhibits MHC class II antigen upregulation, which shows an immunomodulating effect.
Lowers LDL cholesterol in men
Nestel P reported a clinical experiment both for men and women. It is a randomized, placebo-controlled, double-blind trial. A total of 46 middle-aged men and 34 postmenopausal women took part in the study. Each group received in crossover design 40 mg red clover isoflavones, either biochanin or formononetin, or a placebo during 6 weeks. In this study, only the biochanin-rich red clover extract lowered LDL-C and that effect was observed only in men.
Mechanism and metabolism pathway
Experiments in vivo and in vitro estimated Biochanin A partly forms genistein and Fomononetin can be metabolized to Daidzein. All of them activate 4 kinds of important receptors, estrogen receptor? (ER?), estrogen receptor ? (ER?), progesterone receptor (PR) and androgen receptor (AR). The research by V. Beck et conducted a large scale compare experiment to evaluate different isoflavone ingredient effect. Estrogen receptors are related with women premenstral syndrome (PMS), loss in bone mineral density and breast disease. PR and AR were regarded that they influence prostate discease. Their result suggested a potential wide application field. More studies will be continued.
