on an empty stomach. i cap my salts as i find i can't stomach drinking them. 8-10 big caps is a tablespoon. make sure you drink a cup of water with it. or even more.

i think that laying down afterwards for about 30min would be beneficial. time of day? i don't know, but you may want to do it when you don't have to run to the bathroom every 15 min!
