I read in one of the fluconazole-synergy papers where they found the mechanism by which fluconazole becomes synergistic with other antifungals.

Since fluconazole interferes with the candida cell membrane, it allows the other substance to slip into the candida cell (where it wouldn't have a chance before).

So the killer combination seems to be the combination of the membrane-disruptor (fluconazole) combined with the straight antifungal.
