Okay, you definitely need both lids off at the same time.
Pit may have an access lid if it is that large. Sounds like a block tank.
Make sure the pipe is CLEAR from the septic to the Pit.
As to back flow - that can only be Water in the outlet pipe and a small area above the pipe in the pit. Pipe enters the pit near the top. Sounds like the pit is a good distance from the septic tank.

Can't really advise on what to try/do until you have this sorted. As to chemical treatment, may be best to wait until pit starts to refill. Treating empty will only effect the lowest level of the pit.
