It seems to be somewhat age dependent. The young autistic kids often get some gains after just a few rounds but middle aged folks often take a couple of months.
It is hard to distinguish gains from supporting supplements from gains from chelation unless you go off supps for a while.
Most folks get some nice gains early from taking the Cutler recommended supplements for their symptoms. So once you are confident you have a heavy metal problem you want to take the supplements that usually help to offset the ways HMs cause or contribute to your symptoms.
There are a lot of really encouraging 1 year reports from middle aged folks. Before that there seem to be quite a few ups and downs as people go through the process. For example, it is so common for folks to overdo it with doses and stress the endocrine system or something. They then take some time off for testing and tweaking of supplements/meds.
I'm describing the situation with some combo of low & frequent dose DMPS, DMSA and ALA (Cutler), not cilantro or IV! I read too many unpleasant stories about the latter two when I was looking into chelation. I was not willing to roll the dice with them. For example:

The HMs did not get in there overnight. They are not coming out overnight.
