There is no medical test that can be used to diagnose Exfoliative Cheilitis.
No blood test or urine that will give you direct answer: yes, it is, or no it is not.
It is not like pregnancy.
Exfoliative Cheilitis is usually diagnosed by eliminating other possible diagnoses, based on visible symptoms.
If your lips are inflamed, if your lips are peeling, a doctor would usually diagnose you either as Exfoliative Cheilitis, or as Oral Allergy Syndrome.
Both diagnoses may present with the same set of symptoms.
Actually, more often than not, Exfoliative Cheilitis is an allergy to something, more often than not, something people eat.
Oral Allergy Syndrome is usually an allergy to fruit and pollen, and plants.
Exfoliative Cheilitis is usually a manifestation of an allergy or intolerance to just about anything.
Different people are allergic or intolerant to different substances, but the manifestation of that intolerance or that allergy may be very similar, inflammation, death of the upper skin layer, cracked lips, peeling of the upper skin layer, raw skin etc.
It could be a lip balm, a food, toothpaste, cosmetics, cat, dog, pollen, fruit, meat, nuts allergy, gluten intolerance, wheat allergy, milk intolerance, etc.
How would you define a cure for EC?
Different people on this forum would come with different definitions for the Cure of Exfoliative Cheilitis.
There are those that think that the only cure is if you eliminate all major causes, and let the body heal itself.

No one single idea or cure worked for everyone.
You can read about many different ideas from those who got cured:
There are almost 100 people who cured peeling lips, not all of them are cured forever, but at least some of them experienced cure long enough to report it here.
Some of them returned to this forum later to report the problem returned.
Some of them returned to this forum years later to report they are still cured.
Make your own conclusions.
